Dual RNA 3’-end processing of H2A.X messenger RNA maintains DNA damage repair throughout the cell cycle

چکیده
Abstract Phosphorylated H2A.X is a critical chromatin marker of DNA damage repair (DDR) in higher eukaryotes. However, gene expression remains relatively uncharacterised. Replication-dependent (RD) histone genes generate poly(A)- mRNA encoding new histones to package during replication. In contrast, replication-independent (RI) synthesise poly(A)+ throughout the cell cycle, translated into variants that confer specific epigenetic patterns on chromatin. Remarkably H2AFX , H2A.X, hybrid gene, generating both and isoforms. Here we report selective removal either isoform reveals different effects types. some cells, RD generates sufficient for deposition onto DDR associated cells making predominantly require this de novo synthesis, required efficient DDR. This highlights importance differential 3’-end processing maintenance effective

No end yet to messenger RNA 3′ processing!
"The end is in siteY' said the title of a review on mRNA 3' processing that appeared in these pages 10 years ago (Birnstiel et al., 1985). Since then, much progress has been made, but a complete understanding of the biochemistry of the 3'-processing reaction leading to polyadenylated messenger RNAs is still lacking. Here, I summarize recent developments in this field, with special emphasis on t...
متن کاملNuclear RNA - Protein Interactions Messenger RNA Processing
Eucaryotic messenger RNA precursors are processed in nuclear ribonucleoprotein particles (hnRNP). Here recent work on the structure of hnRNP is reviewed, with emphasis on function. Detailed analysis of a specific case, the altered assembly of hnRNP in heatshocked Drosophila and mammalian cells, leads to a general hypothesis linking hnRNP structure and messenger RNA processing. A unifying princi...
متن کاملOxyS small RNA induces cell cycle arrest to allow DNA damage repair
To maintain genome integrity, organisms employ DNA damage response, the underlying principles of which are conserved from bacteria to humans. The bacterial small RNA OxyS of Escherichia coli is induced upon oxidative stress and has been implicated in protecting cells from DNA damage; however, the mechanism by which OxyS confers genome stability remained unknown. Here, we revealed an OxyS-induce...
متن کاملMessenger RNA Degradation: Beginning at the End
The mechanisms responsible for mRNA decay in mammalian cells, and how specific sequence elements accelerate decay, are unknown. Recent work indicates that 'ARE' instability elements recruit the exosome to promote rapid 3'-to-5' degradation of the mRNA.
متن کاملRNA Repair: Damage Control
RNA in a cell is subject to many of the same insults as DNA. RNA damage can induce apoptosis and may be exploited for anti-cancer chemotherapy. It is a surprise, however, to learn that cells may repair RNA damage, suggesting a far greater significance of RNA in genotoxic stress.
